Alterations in glucose turnover following single intravenous injections of epinephrine and glucagon in dogs.

چکیده
The technique of priming injection-continuous infusion of tracer glucose-U-C14 was employed in 7 anesthetized dogs, each serving as its own control, to determine the immediate changes in rates of appearance (Ra) and disappearance (Rd) of plasma glucose following single intravenous injections of epinepbrine (IO pg. per Kg.) and glucagon (I pg. per Kg.). Mean maximal hyperglycemia induced by the hormones was comparable in magnitude, but it was achieved in 4 minutes after epinephrine injection compared with 8 to 10 minutes after glucagon. Furthermore, Ra was increased 5to ‘I-fold during the first 4 minutes following epinephrine injection and subsequently fell to below control levels for about 30 minutes. In contrast, following glucagon, Ra was increased 3to 4-fold during the first 8 minutes after injection with subsequent return to control levels. After both epinephrine and glucagon, Rd increased 1.5to a-fold during the first 10 minutes; it returned to control levels IO minutes after epinephrine and 30 minutes after glucagon. These data indicate that the increment in plasma glucose results from enhanced hepatic glycogenolysis. However, contrary to the suggestion of others, the effect of epinephrine on the liver does not appear to be mediated via glucagon release. (Metabolism 17: No. 7, July, 631-637, 1968)

Posttranscriptional regulation of adrenal TH gene expression contributes to the maladaptive responses triggered by insulin-induced recurrent hypoglycemia
Acute metabolic stress such as insulin-induced hypoglycemia triggers a counterregulatory response during which the release of catecholamines (epinephrine), the activation of tyrosine hydroxylase (TH) enzyme and subsequent compensatory catecholamine biosynthesis occur in the adrenal medulla. However, recurrent exposure to hypoglycemia (RH), a consequence of tight glycemic control in individuals ...
متن کاملAction of hydralazine on glucose turnover and plasma lipids in dogs.
Acute effects of a single intravenous injection of hydralazine (20 mg./Kg.) on heart rate, femoral arterial blood pressure, glucose turnover (technique of priming injection-continuous infusion of glucose-U-C”) and plasma lipids and catecholamines were studied in 15 anesthetized dogs. In 6 of the dogs, 3 received isotonic saline solution and 3 received hydralazine diluent. These control dogs had...
متن کاملSynergistic interactions of physiologic increments of glucagon, epinephrine, and cortisol in the dog: a model for stress-induced hyperglycemia.
To evaluate the role of anti-insulin hormone actions and interactions in the pathogenesis of stress-induced hyperglycemia, the counterregulatory hormones, glucagon, epinephrine, and cortisol were infused alone as well as in double and triple combinations into normal conscious dogs in doses that were designed to simulate changes observed in severe stress. Infusion of glucagon, epinephrine, or co...

متن کاملInteraction of glucagon and epinephrine in the control of hepatic glucose production in the conscious dog.
Epinephrine increases net hepatic glucose output (NHGO) mainly via increased gluconeogenesis, whereas glucagon increases NHGO mainly via increased glycogenolysis. The aim of the present study was to determine how the two hormones interact in controlling glucose production. In 18-h-fasted conscious dogs, a pancreatic clamp initially fixed insulin and glucagon at basal levels, following which one...
